IGF-1 LR3 Facts

What is IGF-1 LR3?

IGF-1 LR3 (Long R3 Insulin-Like Growth Factor-1) is a synthetic analog of human Insulin-Like Growth Factor-1 (IGF-1). It was engineered with two structural modifications: an arginine substitution at position 3 and a 13-amino-acid extension at the N-terminus. These changes reduce its binding to IGF-binding proteins (IGFBPs), allowing it to remain active longer than native IGF-1.

Unlike natural IGF-1, IGF-1 LR3 was developed as a research compound and has not been approved as a therapeutic medication.

Basic Information

  • Compound Class: Synthetic IGF-1 analog
  • Length: 83 amino acids
  • Naturally Occurring: No
  • Primary Target: Insulin-Like Growth Factor-1 Receptor (IGF-1R)
  • Key Modification: Reduced affinity for IGF-binding proteins (IGFBPs)
  • Research Focus:
    • Cell growth
    • Tissue repair
    • Muscle biology
    • Metabolic signaling
    • Growth factor signaling

Mechanism of Action

IGF-1 LR3 binds to the IGF-1 receptor (IGF-1R), activating intracellular signaling pathways involved in cellular growth, survival, and metabolism.

Researchers have found that activation of IGF-1R influences pathways such as:

  • PI3K/Akt
  • mTOR
  • MAPK/ERK

Because IGF-1 LR3 binds less readily to IGF-binding proteins than native IGF-1, a larger fraction remains biologically available, resulting in a longer duration of receptor activation.

Areas of Research

Muscle Biology

Researchers have investigated IGF-1 LR3 for its potential effects on:

  • Muscle cell proliferation
  • Protein synthesis
  • Satellite cell activation
  • Skeletal muscle growth signaling

Most of this evidence comes from laboratory and animal research.

Tissue Repair

Research has explored IGF-1 LR3 for its role in:

  • Connective tissue biology
  • Cellular regeneration
  • Wound healing models
  • Musculoskeletal tissue repair

These findings remain primarily preclinical.

Metabolic Research

IGF-1 signaling is involved in several metabolic processes.

Researchers have investigated IGF-1 LR3 in relation to:

  • Glucose metabolism
  • Nutrient signaling
  • Cellular energy regulation
  • Insulin-related signaling pathways

Additional research is needed to better understand these effects in humans.

Cell Growth Research

IGF-1 LR3 is commonly used in laboratory research involving:

  • Cell proliferation
  • Cell differentiation
  • Growth factor signaling
  • Tissue engineering

Its extended activity makes it useful for studying IGF-1 receptor biology.
